Liam Tack, David Lynn and Thomas Ashworth were on the scoresheet for the hosts, who extended their unbeaten run to six matches, in the Evo-Stik South League encounter.
Tony Burnett netted in stoppage time for Dunstable, but it was merely a consolation.
Thame occupy the final play-off spot and now have a three-point cushion over sixth-placed Welwyn Garden City.
Didcot Town lost on penalties at home to Stratford Town in the third round of the CSS League Challenge Cup.
The tie finished 1-1 after 90 minutes, with Adam Learoyd’s opener for the hosts cancelled out by Jordan Williams.
Stratford then won the shoot-out 4-2 to progress.
